ADCC, antibody-dependent cell-mediated cytotoxicity, refers to the process in which the Fab fragment of an antibody binds to an antigenic epitope on virus-infected cells or tumour cells, while its Fc fragment binds to Fc receptors on effector cells (NK cells, macrophages, etc.), mediating direct killing of the target cells.

Process of Action
▪ IgG antibodies specifically bind to antigenic determinants on the surface of target cells;
▪ Natural killer (NK) cells then bind to the Fc fragment of the IgG on target cells via their corresponding surface receptors;
▪ Activated NK cells release cytotoxic substances such as perforin and granzyme to kill target cells;
▪ The target cells undergo apoptosis, and the antibodies are processed by the liver.

ADCC is currently an important mechanism by which many antibody drugs exert their efficacy. Although genetically engineered cell lines are available, PBMC can better recapitulate the ADCC effect in the human body, making PBMC screening particularly important.
To address the challenge of PBMC screening, Sailybio provides a complete ADCC experimental solution and recommends PBMC batches that perform well in ADCC screening to customers.
