CFDA SE (5(6)-carboxyfluorescein diacetate succinimidyl ester)
freely diffuses across the cell membrane and is converted
intracellularly by esterases into fluorescent CFSE, which covalently
binds to intracellular proteins. With each cell division the CFSE
fluorescence intensity is halved. By detecting the successive peaks
of stepwise decreasing fluorescence intensity through flow cytometry,
both the number of proliferation generations and the proportion of
dividing cells can be accurately tracked - making this the classic
method for assessing proliferative capacity.
