In the presence of complement, Rituximab exhibited marked CDC killing of Raji cells, whereas the CD3 bispecific antibody did not mediate killing of CD3+ T cells.

Complement-Dependent Cytotoxicity (CDC): complement-dependent cytotoxicity occurs when the Fc region of an antibody binds to and activates complement molecules while the Fab region binds to the target cell, leading to formation of the membrane attack complex and ultimately resulting in lysis of the target cell.
Effector cells and target cells are co-incubated at a defined effector-to-target (E:T) ratio, and the CDC effect is measured under stimulation with antibody drugs.
SailyBio provides CDC (complement-dependent cytotoxicity) assay services: using Rituximab-mediated CDC killing as a validation case, the assay measures complement activation via the antibody Fc region and the resulting target cell lysis, supporting complement killing assays and antibody efficacy evaluation, and meeting the CDC functional verification requirements of monoclonal and bispecific antibodies.
